Short hairpin RNA (shRNA) is an RNA molecule delivered via vectors and stably expressed in cells, which can be processed into small interfering RNA (siRNA) to guide the RNA-induced silencing complex (RISC) in mediating the degradation of target messenger RNA (mRNA), thereby achieving specific gene knockdown. A single shRNA experiment can only investigate the function of one specific gene. However, most biological phenomena—such as cell proliferation, death, differentiation, carcinogenesis, and drug response—are not determined by a single gene but are co-regulated by complex gene networks.
